About Tesamorelin 10mg (Single Vial)

Tesamorelin is a stabilised analogue of growth hormone-releasing hormone (GHRH). A trans-3-hexenoyl group attached to the N-terminus protects it from rapid enzymatic degradation, giving it a longer active window than native GHRH while preserving receptor binding.

Because it acts upstream — stimulating the pituitary to release its own growth hormone rather than supplying growth hormone directly — it is studied as a secretagogue, and the resulting release follows the body's own pulsatile pattern.

Research background

GHRH receptor agonism

Tesamorelin binds pituitary GHRH receptors, stimulating endogenous growth hormone release. The distinction from exogenous growth hormone administration is that feedback regulation via somatostatin remains intact.

Visceral adipose tissue

The most substantial clinical literature concerns visceral adipose tissue measured by CT imaging, examined in populations with HIV-associated lipodystrophy. This is the indication for which it received approval.

Hepatic fat and liver enzymes

A related strand examines liver fat content and enzyme markers alongside visceral fat reduction, addressing whether the two move together.

IGF-1 as a downstream marker

Studies routinely track insulin-like growth factor 1 as the measurable downstream consequence of growth hormone release, since IGF-1 has a longer half-life and is easier to assay than pulsatile GH itself.

Reconstitution

  1. Equilibrate firstAllow the vial to reach room temperature before opening. Introducing solvent into a cold vial promotes condensation, which affects both concentration accuracy and stability.
  2. Select the diluentBacteriostatic water — sterile water with 0.9% benzyl alcohol — is standard where the solution will be held for more than a few days. Plain sterile water is appropriate only for single-session use with no storage.
  3. Add slowly, down the wallDirect the solvent against the inner wall of the vial rather than onto the powder. A stream striking the lyophilized cake causes localized shear that can denature peptide chains.
  4. Do not shakeSwirl gently, or simply let the vial stand. Agitation introduces air and mechanical stress, and both degrade peptides. Full dissolution usually takes a minute or two.
  5. Inspect before useA correctly reconstituted solution is clear and colourless. Cloudiness, visible particulate or discolouration indicates a problem with the material or the diluent.

Concentration is set by the volume of diluent added. For a 10mg vial, 2mL of bacteriostatic water yields 5mg/mL. Calculate the working concentration your protocol requires rather than following a generic figure.

Lyophilized peptide is stable enough to survive shipping at ambient temperature, which is why cold-chain courier service is not required for transit. Long-term storage is a different matter — material intended to be held for months belongs in a freezer.

Protect from light, and avoid repeated freeze-thaw cycles. Each cycle causes measurable degradation, so dividing a reconstituted stock into single-use aliquots before freezing is preferable to thawing and refreezing one vial repeatedly.

Purity and verification

Every lot is tested by an independent laboratory using two orthogonal methods. HPLC (high-performance liquid chromatography) establishes purity as a percentage of total peptide content, separating the target compound from truncated sequences and synthesis by-products. LC-MS (liquid chromatography–mass spectrometry) confirms molecular identity by mass.

Both matter. Purity alone is insufficient — a 99% pure sample of the wrong compound is still the wrong compound. Together the two tests establish identity and quality, which is why Tesamorelin is released only after both pass.

How do you reconstitute tesamorelin?
Bring to room temperature, add bacteriostatic water slowly down the vial wall, swirl gently. A 10mg vial with 2mL gives 5mg/mL. GHRH analogues are particularly sensitive to agitation, so avoid shaking entirely.
How does tesamorelin differ from growth hormone?
Tesamorelin stimulates the pituitary to release its own growth hormone; it is not growth hormone. The resulting release is pulsatile and remains subject to the body's own feedback control, which is the main pharmacological distinction studied in the literature.
How does it differ from sermorelin or ipamorelin?
Sermorelin is a shorter GHRH fragment with a much shorter half-life. Ipamorelin acts on a different receptor entirely — the ghrelin/GHS receptor — rather than the GHRH receptor. Tesamorelin is the stabilised GHRH analogue with the largest clinical dataset.
